中文摘要:目的 建立同时测定谷光甘肽原料及制剂中氧化型和还原型谷光甘肽的反相高效液相色谱法.方法 采用外标法,以Hypersil C18为色谱柱,25 mmol/L KH2PO4(pH2.5)∶四氢呋喃(100∶1)为流动相,流速为1.0 ml/min,柱温为30℃,检测波长230 nm,进行分离测定.结果 在3.12~2 490.0μg/ml间,GSH线性关系良好(r=0.9999),测得回收率为101.84%.在3.25~975.0 |

Determination of glutathione and oxidized glutathione by HPLC |

Abstract:Objective To establish a method to determine GSH and GSSG by HPLC. Method The analytical column was Hypersil C18. The mobile phase consisted of 25m mol/L KH2PO4 (pH2.5):tetrahydrofuran (100:1). The flow rate was 1.0 ml/min. Column temperature was 30 ℃ and the detective wavelength was 230 nm. Result The calibration curve of GSH was linear within the range of 3.12 μg/ml~2490.0 μg/ml (r=0.9999) and the average recovery was 101.84%. The calibration curve of GSSG was linear within the range of 3.25 μg/ml~975.0 μg/ml (r=0.9999) and the average recovery was 97,84%. Conclusion The method was proved to be reliable and accurate,which could be used to determinate GSH and GSSG simultaneously. |
