中文摘要:目的 建立HPLC法测定奥卡西平活性代谢产物单羟基卡马西平的血药浓度。方法 血浆样品经甲醇处理后,以奥硝唑为内标,采用HPLC法测定,其中,色谱柱为ZORBAX Eclipse XDB-C18(150 mm×4.6 mm,5 μm),流动相为水-乙腈(80∶20,V/V),流速1.0 ml/min,柱温35 ℃,双波长检测:192 nm检测单羟基卡马西平,318 nm检测奥硝唑。结果 单羟基卡马西平血药浓度在2~50 μg/ml(r=0.998 6)范围内线性关系良好,定量下限浓度为2 μg/ml;准确度在95.57%~100.59%之间,日内、日间精密度RSD均小于15%;单羟基卡马西平及内标的平均提取回收率在89.62%~98.76%之间;稳定性的RSD均小于6%。结论 该方法专属性强、灵敏度高,且操作简便,可用于单羟基卡马西平血药浓度的临床检测。

The assay of monohydroxy carbamazepine, the active metabolite of oxcarbazepine, in human plasma by HPLC
Abstract:Objective To establish an assay method for monohydroxy carbamazepine, the active metabolite of oxcarbazepine, in human plasma.Methods Ornidazole was used as the internal standard. Plasma samples were processed with methanol and analyzed by HPLC. The column was ZORBAX Eclipse XDB-C18(150 mm×4.6 mm, 5 μm) with the mobile phase of water-acetonitrile (80∶20, V/V) at a flow rate of 1.0 ml/min. Dual wavelength detection is applied. The detection wavelength of monohydroxy carbamazepine was set at 192 nm and ornidazole at 318 nm.Results There was an excellent liner relationship for monohydroxy carbamazepine from 2 to 50 μg/ml(r= 0.998 6). The limit of quantification was 2 μg/ml with the range of accuracy between 95.57% and 100.59%. The RSD of intra-day and inter-day precisions were less than 15%. The average extraction recovery rate of MHC and internal standard were in the range of 89.62% to 98.76%. The RSD of stability was less than 6%.Conclusion This method is specific, sensitive, and easy to operate. It is suitable for the clinical assay of monohydroxy carbamazepine in human plasma.
